Identification of Mycobacterium leprae-induced cellular proteins in IC21 macrophage cell

Abstract
In order to survive intracellularly, Mycobacterium leprae, an obligatory parasite, should
be able to counteract host cell¡¯s antimicrobial activities and/or modify host cellular
environment. Toward understanding of such intracellular modification by M. leprae, we
haute investigated the changes of protein expression profiles of macrophage in response
to M. leprae infection by 2-D gel electrophoresis analyses. Proteins of IC21 cells, a
macrophage cell line, infected with viable M. leprae and heat-killed M. leprae were
labeled with [35S]-methionine. The metabolically labeled proteins were
resolved by two-dimensional gel electrophoresis and analyzed quantitatively using a
phosphorimage analyzer. It was found that infection with M. leprae induced a significant
change in the gene expression profile of IC21 macrophage cell. The expression of at
least 19 macrophage proteins was induced and that of 4 proteins was repressed by
viable M. leprae infection. Such proteins might be important in the protective immune
responses to M. leprae infection and/or they might be related with potential virulence
determinants of M. leprae. Identification and characterization of M. leprae-induced
proteins in macrophage could provide valuable informations to our understanding of
pathogenesis of leprosy.
